2010 TE250 wont start, got spark, fuel, good timing and compression

Hi, I've had this bike for a while and it randomly stopped one day. so matter what i try i cant get it to start. So far I've, got a new spark plug and am getting great spark, have cleaned and checked everything in the throttle body and the injector in working perfectly, done a full fuel flush through with new good fuel and checked the pressure of the fuel pump which is all fine. Checked my valve timing and clearances specs which is all within limits.i tested the temperature sensor problem and the electric starter issue to see if im getting them and they are working perfect with no issues. I've cleaned and checked the exhaust, done a full oil change and i still cant get it to start. there are probably more things that i haven't mentioned that I've forgotten. Im getting fuel, spark, timing is good and i have good compression. the only thing ive been able to do is make it backfire a fair bit and blow the throttle body of the clamp to the head. Im totally stuck so any help would be awesome.

When I hear backfire through the intake, i think out of time.....

Sounds like its out of time to me. If you actually had all of those things correct it would run. Triple check it all.

just totally rechecked the timing its all perfect, any ideas?

Have you checked the little key that keeps the flywheel in place? If that is broken the timing will be off even if the chain/valves are ok.
